Gabimaru and the others come across what seems like an empty and abandoned village. Suddenly Gabimaru senses someone behind them. Gabimaru looks behind and finds a little girl running away.
As Gabimaru chases her, he is attacked by another creature who looks like a tree. However, he and Sagiri continue their chase and leave the creature to Senta and Yuzuriha. As they catch the girl, she shows resistance and fights the pair. Even though she was exceptionally powerful, she still was no match for Gabimaru.
Sagiri and Gabimaru are left speechless when the little powerful girl starts crying as Gabimaru takes her hostage. Sagiri then asks Gabimaru to untie and comfort her. The pair start to go back to reunite with Senta and Yuzuriha.
The tree-like creature was defeated and asked Gabimaru to return the little girl, and he would tell them everything he knows about the island. The creature introduces the girl as Mei and himself as Hoko.

Hoko takes the squad to his house and provides them with some food and a bath. He then proceeds to tell the squad that he has lived in the village for 1,000 years. And the Elixir of Life that they are looking for is actually called Tan. Gabimaru then asks him where it is.
Hoko tells Gabimaru that the island is divided into 3 regions, Eishu, Hojo, and Horai. The outermost region where the beach and the woods are is Eishu, the village they are in is Hojo, and the Tan is somewhere in Horai, which is the middle of the Island.
The Three Regions Of Kotaku/Shinsenkyo

These are the regions that Shinsenkyo Island is divided into:
Eishu
Eishu, situated on the outer edge of the island, is characterized by its dense forest and stunning beach. The forest, which is teeming with plant life, owes its verdant appearance to the decomposing remains of samurai who dared to venture onto the island in the past.
Despite the allure of Eishu's natural beauty, the area is fiercely guarded by the Soshin, whose duty is to protect the island from any intruders. Their presence in Eishu serves as a warning to any who would consider trespassing on the Island.
Hojo
Hojo is the second region of the island, consisting of a valley that encircles Horai and serves as the home base for the Soshin guardians. A millennium ago, the Hoko race inhabited a small village in Hojo until they underwent Arborification, resulting in the village being abandoned and left in ruins.
In the present day, only two individuals, Hoko and Mei, are known to reside in the deserted village. This is the place that Gabimaru and the others discover after defeating Rokurota, the Giant of Bizen.
Horai
At the heart of the island lies Horai, a place of great significance, as it is where the powerful Lord Tensen and the skilled Doshi reside. Lord Tensen are the powerful beings who have mastered the practices of Immortality.
Meanwhile, Doshi serves as their students, servants, and army to protect Lord Tensen. Tan is somewhere within Horai, according to Hoko, but Horai is covered in mist, and the path to Horai is really tough.
